Storage	Capture Antibody: 360 ug ofantigen-affinity purified goat anti-IL-6. Centrifuge vial prior to opening. Reconstitutein 1.0 ml PBS for a concentration of 360 ug/ml. Following reconstitution theCapture antibodies may be stored at 2-8C for up to 6 months. For long term storage it isrecommended to aliquot into working volumes and store at -70Cin a manual defrostfreezer. Detection Antibody: 36 ug ofbiotinylated antigen-affinity purified goat anti-IL-6. Centrifuge vial priorto opening. Reconstitute in 1.0 ml sterile 1.0% BSA in PBS for aconcentration of 36 ug/ml. Following reconstitution the Detection antibodiesmay be stored at 2-8C for upto 6 months. For long term storage it is recommended to aliquot into workingvolumes and store at -70C in amanual defrost freezer. Human IL-6 Standard: 17.5 ngof recombinant IL-6. Centrifuge vial prior to opening. Reconstitute in 0.5 mlsterile water for a concentration of 35 ng/ml. The Standard may be stored at2-8C for onemonth or aliquoted and stored at -70C for up to three months in a manual defrost freezer. UltraAvidin-HRPConjugate: 40 ulvial. Upon receipt, UltraAvidin-HRP conjugate should be stored at 2-8C, DO NOT FREEZE. TMB LiquidSubstrate: Aspirate and washplate 4 times. Add 100 ul of TMB HRPO Microwell Substrate Standard KineticOne Component Ready Use to each well. Incubate at roomtemperature and monitorcolor development with an ELISA plate reader at 450 nmwith wavelength correction set at 540 nm or 570 nm. The reaction may bestopped after 15-20 minutes by adding 100 ul of 2 M sulfuric acid to eachwell.
Intended Use	RUO, Human IL-6 ELISAdevelopment kit contains the key components required for the quantitative measurementof natural and/or recombinant IL-6 in a sandwich ELISA format within the rangeof 32-2, 000 pg/ml. Using the ELISA protocol described below, this kitprovides sufficient reagents to assay IL-6 in approximately 1, 500 ELISA platewells.

ELISA Procedure	Standard/Sample: Dilutestandard from 2, 000 pg/ml to zero in diluent. Immediately add 100 ul ofstandard or sample to each well in duplicate and incubate at room temperaturefor at least 2 hours. Detection: Aspirateand wash plate 4 times. Dilute a portion of detection antibody in diluent toa concentration of 50 ng/ml. Add 100 ul per well and incubate at room temperaturefor 2 hours. UltraAvidin-HRP Conjugate: Aspirate and wash plate 4 times. Dilute 3.0 ul of avidin-HRPconjugate 1:5, 000 (this dilution factor may require some optimization) indiluent for a total volume of 15 ml. Add 100ulper well and incubate at room temperature for 30 minutes. TMB Liquid Substrate:Add 100 ul of TMB HRPO Microwell Substrate StandardKinetic One Component Ready Use to each well. Incubate at roomtemperature for 20-30 minutes and monitor color development with an ELISAplate reader at 450 nm with wavelength correction set at 540 nm or 570 nm.Avoid placing plates in direct light. Stop Solution: The reactionmay be stopped after 15-20 minutes by adding 100 ul of 2 M sulfuric acid toeach well. All eye, hand, face, and clothing protection should be used when workingwith sulfuric acid.
