Purified Recombinant Human CD30 (Accession # P28908)
This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.3 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.
The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

Flow Cytometry: It is recommended to use the indirect method for signal enhancement when enumerating cells expressing CD30. A suggested method would be to stain cells expressing CD30 with 0.25 µg/106 cells in a total staining volume of ≤200 µl followed by PN:M1189.Western Blotting: To detect Human CD30 this monoclonal antibody can be used at a concentration of 1-2 µg/ml. This monoclonal antibody should be used in conjunction with compatible second-step reagents such as PN:M114 and a chromogenic substrate such as PN:T343. The detection limit for Human CD30 is 10 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used. A suitable Western blotting control is PN:C1357.Functional Assay: The agonist activity of anti-human CD30 was measured by its ability to stimulate human IL-6 secretion by HDLM-2 cells (Hodgkin’s lymphoma cell line) (Blood, 1996, 87(6):2443 - 2449). The ED50 of this effect is typically 0.05-0.2 μg/mL.

Mouse Anti-Human CD30 (Clone 81337) recognizes an epitope on Human CD30. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.
CD30/TNFRSF8, a type I transmembrane glycoprotein belonging to the TNF receptor superfamily and tumor marker. This receptor is expressed by activated, but not by resting, T and B cells. TRAF2 and TRAF5 can interact with this receptor, and mediate the signal transduction that leads to the activation of NF-kappaB. It is a positive regulator of apoptosis, and also has been shown to limit the proliferative potential of autoreactive CD8 effector T cells and protect the body against autoimmunity. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. CD30 is associated with anaplastic large cell lymphoma. It is expressed in embryonal carcinoma but not in seminoma and is thus is a useful marker in distinguishing between these germ cell tumors.
