Purified Recombinant Mouse B7-2 (> 98%)
This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.3 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.
The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

Indirect ELISA: This antibody can be used in an indirect detection ELISA at 0.5 - 1.0 µg/ml with a suitable second step reagent such as PN:R380. The detection sensitivity of this indirect ELISA for Mouse B7-2 is approximately 1 ng/well.Flow Cytometry: It is recommended to use the indirect method for signal enhancement when enumerating cells expressing B7-2. A suggested method would be to stain cells expressing B7-2 with 0.25 µg per 1 - 2.5 x 106 cells in a total staining volume of ≤200 µl followed by PN:R485.Western Blotting: To detect Mouse B7-2 this monoclonal antibody can be used at a concentration of 1-2 µg/ml. This monoclonal antibody should be used in conjunction with compatible second-step reagents such as PN:R377 and a chromogenic substrate such as PN:T343. The detection limit for rmB7-2 is approximately 0.1 ng/lane and 0.5 ng/lane under non-reducing and reducing conditions, respectively. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
Neutralization: This antibody is useful for neutralization of Mouse B7-2 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Mouse B7-2 (at 10 ng/ml) is 2.5 - 10 µg/ml.CyTOF-ready:Ready to be labeled using established conjugation methods. No BSA or other carrier proteins that could interfere with conjugation.
Clone GL-1 recognizes an epitope on mouse CD86.
CD86 is expressed on activated B and T cells, macrophages, dendritic cells, and astrocytes.
CD86 is an 80kD Ig superfamily member that is involved in immunoglobulin class-switching and activation of NK cell-mediated cytotoxicity. CD80 is closely related to, and works in tandem with CD86 to prime T- cells. CD86 is expressed earlier in the immune response than CD80. The ligation of CD28 on T cells with CD80 and CD86 on APCs co-stimulates T cells resulting in enhanced cell activation, proliferation, and cytokine production. CD86 can also bind to CTLA-4 to deliver an inhibitory signal to T cells.
