Specificity:
This antibody reacts with a 110 kD human cell surface glycoprotein known as HNK-1. This CD marker recognizes peripheral blood lymphocyte subsets and natural killer (NK) cells. Many cells which co-express CD57 and CD8 antigens are a subset of suppressor/cytotoxic T-cells. These antibodies recognize neuroendocrine cells and their tumors. CD57 antigens are not expressed on erythrocytes or platelets.

Application Note:
Immunofluorescence 5-20 ug/mL. Flow cytometry 1-5 ug/10^6 cells. Immunoblotting 1-5 ug/mL. This antibody can be used on frozen cryostat sections as well as formalin-fixed paraffin-embedded tissue sections. Prolonged fixation is buffered formalin can destroy the epitope. The antibody may be used at a dilution of 5-10 ug/mL with the Auto/Probe III kit. For consistent and reproducible results on formalin-fixed paraffin-embedded sections a high temperature antigen unmasking technique is required (see instructions below). Function:
Involved in the biosynthesis of L2/HNK-1 carbohydrate epitope on glycoproteins. Can also play a role in glycosaminoglycan biosynthesis. Substrates include asialo-orosomucoid (ASOR) asialo-fetuin and asialo-neural cell adhesion molecule. Requires sphingomyelin for activity: stearoyl-sphingomyelin was the most effective followed by palmitoyl-sphingomyelin and lignoceroyl-sphingomyelin. Activity was demonstrated only for sphingomyelin with a saturated fatty acid and not for that with an unsaturated fatty acid regardless of the length of the acyl group (By similarity).
Catalytic Activity:
UDP-glucuronate + 3-beta-D-galactosyl-4-beta-D-galactosyl-O-beta-D-xylosylprotein = UDP + 3-beta-D-glucuronosyl-3-beta-D-galactosyl-4-beta-D-galactosyl-O-beta-D-xylosylprotein. Cofactor: Manganese.
