Interleukin 8

Interleukin-8 (IL-8) is a chemokine produced by macrophagesand other cell types such as epithelial cells. It is also synthesized byendothelial cells, which store IL-8 in their storage vesicles, theWeibel-Palade bodies. In humans, the interleukin-8 protein is encoded by theIL8 gene. There are more receptors of the surface membrane capable to bindIL-8, the most frequently studied types are the G protein coupled serpentinereceptors CXCR1 and CXCR2. Expression and affinity to IL-8 is different inthe two receptors (CXCR1 > CXCR2). Toll-like receptors are the receptorsof the innate immune system.

Purified, Lyophilized. Reconstitute using sterile deionized water to a concentration >=100ug/ml.Further dilutions can be made in other aqueous buffers.

Specificactivity of Human IL-8 determined in chemotaxis of donor PBL neutrophils withthreshold concentration corresponding to 25–150ng/ml. Each laboratory shoulddetermine an optimum working titer for use in its particular application.Other applications have not been tested but use in such assays should not necessarilybe excluded.

Thelyophilized product, though stable for 3 weeks at room temperature, is beststored at -20oC. After reconstitution, short term (up to 1 week) store at2–8oC. Long term, add 0.1% HSA or BSA, aliquot and store at -20oC. Avoidmultiple freeze/thaw cycles.
