Background
Short transient receptor potential channel 3 (TrpC3) also known as transient receptor protein 3 (TRP-3) is a protein that in humans is encoded by the TRPC3 gene. The protein encoded by this gene is a membrane protein that can form a non-selective channel permeable to calcium and other cations. The encoded protein appears to be induced to form channels by a receptor tyrosine kinase-activated phosphatidylinositol second messenger system and also by depletion of intracellular calcium stores. Two transcript variants encoding different isoforms have been found for this gene.
Gene Name
TRPC3
Application Details
Western blot, 0.1-0.5μg/ml, Human, Mouse, Rat
Immunohistochemistry (Paraffin-embedded Section), 0.5-1μg/ml, Human, Mouse, Rat, By Heat
Application Notes
Tested Species: In-house tested species with positive results. Predicted Species: Species predicted to be fit for the product based on sequence similarities. By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections. Other applications have not been tested. Optimal dilutions should be determined by end users.
Predicted Reactivity
Hamster
Clonality
Polyclonal
Concentration
Adding 0.2 ml of distilled water will yield a concentration of 500 μg/ml.
Contents
Each vial contains 5mg BSA, 0.9mg NaCl, 0.2mg Na2HPO4, 0.05mg Thimerosal, 0.05mg NaN3.
Immunogen
A synthetic peptide corresponding to a sequence at the C-terminus of human TRPC3, identical to the related rat and mouse sequences.
Purification
Immunogen affinity purified.
Cross-reactivity
No cross-reactivity with other proteins
Storage
Store at -20˚C for one year from date of receipt. After reconstitution, at 4˚C for one month. It can also be aliquotted and stored frozen at -20˚C for six months. Avoid repeated freeze-thaw cycles.
Reconstitution
Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Gene Full Name
Short transient receptor potential channel 3
Molecular Weight
96009 MW
Protein Function
Thought to form a receptor-activated non-selective calcium permeant cation channel. Probably is operated by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases or G-protein coupled receptors. Activated by diacylglycerol (DAG) in a membrane-delimited fashion, independently of protein kinase C, and by inositol 1, 4, 5- triphosphate receptors (ITPR) with bound IP3. May also be activated by internal calcium store depletion..
Subcellular Localization
Membrane; Multi-pass membrane protein.
Tissue Specificity
Expressed predominantly in brain and at much lower levels in ovary, colon, small intestine, lung, prostate, placenta and testis.
Protein Name
Short transient receptor potential channel 3(TrpC3)
Sequence Similarities
Belongs to the transient receptor (TC 1.A.4) family. STrpC subfamily. TRPC3 sub-subfamily.
